简介:Thisstudyaimedtoexploretheroleofmechanicaltensioninhypertrophicscarsandthechangeinnervedensityusinghematoxylin-eosinstainingandS100immunohistochemistry,andtoobservetheexpressionofnervegrowthfactorbywesternblotanalysis.Theresultsdemonstratedthatmechanicaltensioncontributedtotheformationofahyperplasticscarinthebackskinofrats,inconjunctionwithincreasesinbothnervedensityandnervegrowthfactorexpressioninthescartissue.Theseexperimentalfindingsindicatethatthecutaneousnervoussystemplaysaroleinhypertrophicscarformationcausedbymechanicaltension.
简介:Ciliaryneurotrophicfactoristheonlyknownneurotrophicfactorthatcanpromotedifferentiationofhippocampalneuralprogenitorcellstoglialcellsandneuronsinadultrats.Thisprocessissimilartospontaneousdifferentiation.Therefore,ciliaryneurotrophicfactormaybeinvolvedinspontaneousdifferentiationofneuralstemcells.Toverifythishypothesis,thepresentstudyisolatedneuralprogenitorcellsfromadultmaleratsandculturedtheminvitro.Resultsshowedthatwhenneuralprogenitorcellswereculturedintheabsenceofmitogenfibroblastgrowthfactor-2orepidermalgrowthfactor,theyunderwentspontaneousdifferentiationintoneuronsandglialcells.Westernblotandimmunocytochemicalstainingshowedthatexogenousciliaryneurotrophicfactorstronglyinducedadulthippocampalprogenitorcellstodifferentiateintoneuronsandglialcells.Moreover,passage4adulthippocampalprogenitorcellsexpressedhighlevelsofendogenousciliaryneurotrophicfactor,andaneutralizingantibodyagainstciliaryneurotrophicfactorpreventedthespontaneousneuronalandglialdifferentiationofadulthippocampalprogenitorcells.Theseresultssuggestthatthespontaneousdifferentiationofadulthippocampalprogenitorcellsismediatedpartiallybyendogenousciliaryneurotrophicfactor.
简介:Objective:Thepredictiveandprognosticroleofprognosticnutritionalindex(PNI)ingastriccancerpatientswithperitonealdisseminationremainsunclear.ThisstudyaimstoexploretheroleofthePNIinpredictingoutcomesofgastriccancerpatientswithperitonealdissemination.Methods:Atotalof660patientsdiagnosedwithgastricadenocarcinomawithperitonealmetastasisbetweenJanuary2000andApril2014atSunYat-senUniversityCancerCenterandtheSixthAffiliatedHospitalofSunYatsenUniversitywereretrospectivelyanalyzed.Theclinicopathologiccharacteristicsandclinicaloutcomesofpatientswithperitonealdisseminationwereanalyzed.Results:ComparedwithPNI-highgroup,PNI-lowgroupwascorrelatedwithadvancedage(P=0.036),worseperformancestatus(P<0.001),higherfrequencyofascites(P<0.001)andhigherfrequencyofmultisitedistantmetastasis(P<0.001).Kaplan-MeiersurvivalcurvesshowedthatPNI-highgrouphadasignificantlylongermedianoverallsurvivalthanPNI-lowgroup(13.13vs.9.03months,P<0.001).MultivariatesurvivalanalysisrevealedthatBorrmanntypeIV(P=0.014),presenceofascites(P=0.017)andlowerPNI(P=0.041)wereindependentpoorprognosticfactors,andpalliativesurgery(P<0.001)andfirst-linechemotherapy(P<0.001)weregoodprognosticfactors.Forpatientsreceivingpalliativesurgery,thepostoperativemorbidityratesinthePNI-lowgroupandPNIhighgroupwere9.1%and9.9%,respectively(P=0.797).ThepostoperativemortalityratewasnotsignificantlydifferentbetweenPNI-lowandPNI-highgroups(2.3%vs.0.9%,P=0.362).Conclusions:PNIisausefulandpracticaltoolforevaluatingthenutritionalstatusofgastriccancerpatientswithperitonealdissemination,andisanindependentprognosticfactorforthesepatients.
简介:Asecond-ordermixingdifferenceschemewithalimitingfactorisdeducedwiththereconstructiongradientmethodandappliedtodiscretizingtheNavier-Stokesequationinanunstructuredgrid.Thetransformofnonorthogonaldiffusionitemsgeneratedbytheschemeindiscreteequationsisprovided.TheDelaunaytriangulationmethodisimprovedtogeneratetheunstructuredgrid.ThecomputingprogrambasedontheSIMPLEalgorithminanunstructuredgridiscompiledandusedtosolvethediscreteequationsoftwotypesofincompressibleviscousflow.Thenumericalsimulationresultsofthelaminarflowdrivenbylidincavityandflowbehindacylinderarecomparedwiththetheoreticalsolutionandexperimentaldatarespectively.Intheformercase,agoodagreementisachievedinthemainvelocityanddragcoefficientcurve.Inthelattercase,thenumericalstructureanddevelopmentofvortexunderseveralReynoldsnumbersmatchwellwiththatoftheexperiment.Itisindicatedthatthefactordifferenceschemeisofhigheraccuracy,andfeasibletobeappliedtoNavier-Stokesequation.
简介:Transcriptionfactor(TF)bindingtoitsDNAtargetsiteplaysanessentialroleingeneregulation.Thelocation,orientationandspacingoftranscriptionfactorbindingsites(TFBSs)alsoaffectregulatoryfunctionoftheTF.However,hownucleosomalcontextofTFBSsinfluencesTFbindingandsubsequentgeneregulationremainstobeelucidated.Usinggenome-widenucleosomepositioningandTFbindingdatainbuddingyeast,wefoundthatbindingaffinitiesofTFstoDNAtendtodecreasewithincreasingnucleosomeoccupancyoftheassociatedbindingsites.WefurtherdemonstratedthatnucleosomalcontextofbindingsitesiscorrelatedwithgeneregulationofthecorrespondingTF.Nucleosome-depletedTFBSsarelinkedtohighgeneactivityandlowexpressionnoise,whereasnucleosome-coveredTFBSsareassociatedwithlowgeneactivityandhighexpressionnoise.Moreover,nucleosome-coveredTFBSstendtodisruptcoexpressionofthecorrespondingTFtargetgenes.WeconcludethatnucleosomalcontextofbindingsitesinfluencesTFbindingaffinity,subsequentlyaffectingtheregulationofTFsontheirtargetgenes.ThisemphasizestheneedtoincludenucleosomalcontextofTFBSsinmodelinggeneregulation.
简介:Therearemanyfactorsaffectingtheinstabilityofthesubmarinehydrate-bearingslope(SHBS),andtheinteractionwithhydrateisverycomplicated.Inthispaper,themechanicalmechanismofthestaticliquefactionandinstabilityofsubmarineslopecausedbythedissociationofnaturalgashydrate(NGH)resultingintherapidincreaseofporepressureofgashydrate-bearingsediments(GHBS)andthedecreaseofeffectivestressareanalyzedbasedonthetimeseriesandtypeofSHBS.Then,takingthetypicalsubmarineslopeinthenorthernSouthChinaSeaasanexample,fourimportantfactorsaffectingthestabilityofSHBSareselected,suchasthedegreeofhydratedissociation,thedepthofhydrateburial,thethicknessofhydrate,andthedepthofseawater.Accordingtotheprincipleoforthogonalmethod,25orthogonaltestschemeswith4factorsand5levelsaredesignedandthesafetyfactorsofsubmarineslopestabilityofeachschemearecalculatedbyusingthestrengthreductionfiniteelementmethod.Bymeansoftheorthogonaldesignrangeanalysisandthevarianceanalysis,sensitivityofinfluentialfactorsonstabilityofSHBSareobtained.Theresultsshowthatthedegreeofhydratedissociationisthemostsensitive,followedbyhydrateburialdepth,thethicknessofhydrateandthedepthofseawater.Finally,theconceptofgashydratecriticalburialdepthisputforwardaccordingtotheinfluencelawofgashydrateburialdepth,andthenumericalsimulationforspecificsubmarineslopeiscarriedout,whichindicatestheexistenceofcriticalburialdepth.

简介:Toexplorethemolecularmechanismoftheprotectiveeffectofnervegrowthfactor(NGF)oninjuredspinalcord.Methods:TheposteriorT8(the8ththoracicsegment)spinalcordsof60Wistarratswereinjuredbyimpactscausedbyobjects(weighing10g)fallingfromaheightof2.5cmwithAllensway.Solutionwithnervegrowthfactors(NGF)wasgivento30rats(theNGFgroup)throughamicrotubuleinsertedintothesubarachnoidcavityimmediately,andat2,4,8,12and24hoursafterspinalcordinjury(SCI)respectively.Normalsaline(NS)withsamevolumewasgiventotheother30rats(theNSgroup)withthesamemethod.And5normalratsweretakenasthenormalcontrols.Theexpressionofbcl-2andbaxproteinsinspinalcordwasdetectedwithimmunohistochemistry.Theapoptoticneuronsinspinalcordweremeasuredwithterminaldeoxynucleotidyltransferase-mediateddUTP-biotinnickend-labelingofDNAfragments(TUNEL)staining.Results:Thepositiveexpressionofbcl-2proteinwasstronginthenormalcontrols,butdecreasedintheNSgroup,andincreasedsignificantlyintheNGFgroupascomparedwiththatoftheNSgroup(P<0.01).Thepositiveexpressionofbaxproteinwasalsostronginthenormalcontrols,butincreasedintheNSgroup,anddecreasedsignificantlyintheNGFgroupascomparedwiththatoftheNSgroup(P<0.01).ApoptoticneuronswerefoundintheNSgroup,andtheydecreasedsignificantlyintheNGFgroupascomparedwiththatoftheNSgroup(P<0.01).Conclusions:NGFcanprotecttheinjurednervetissuesthroughstimulatingtheexpressionofbcl-2protein,inhibitingtheexpressionofbaxproteinandinhibitingtheneuronalapoptosisafterSCI.

简介:Thestressintensityfactor(SIF)isacriticalparameterassociatedwiththefracturebehaviourofmaterials.Inthispaper,weselectthedisplacementfunctionaroundacracktipastheshapefunctionofthedigitalimagecorrelation(DIC),whichmakesitpossibletodirectlycalculatetheSIFbythecorrelationscheme.Moreover,weuseanon-rectangularsubset,whichcanreducetheinfluenceofplasticdeformationandcrackwidthontheDICmeasurementaccuracy.WemeasuredtheSIFofamodeIcrackinasuper-hardaluminiumalloyspecimentoverifytheperformanceoftheproposedmethod.OurexperimentalresultsshowthataDICwithaspecificshapefunctioncanbeusedtoaccuratelyandefficientlycalculatetheSIF.Furthermore,wealsopresentapracticalapplicationofourproposedmethodfordeterminingtheSIF,crackpropagationangleandcracktipdisplacement.
简介:Gastrodin,anactivecomponentoftallgastrodiatuber,iswidelyusedinthetreatmentofdizziness,paralysis,epilepsy,strokeanddementia,andexhibitsaneuroprotectiveeffect.AratmodelofspinalcordinjurywasestablishedusingAllen’smethod,andgastrodinwasadministeredviathesubarachnoidcavityandbyintraperitonealinjectionfor7days.Resultsshowthatgastrodinpromotedthesecretionofbrain-derivedneurotrophicfactorinratswithspinalcordinjury.Aftergastrodintreatment,themaximumangleoftheinclinedplanetest,andtheBasso,BeattieandBresnahanscoresincreased.Moreover,gastrodinimprovedneuraltissuerecoveryintheinjuredspinalcord.Theseresultsdemonstratethatgastrodinpromotesthesecretionofbrain-derivedneurotrophicfactor,contributestotherecoveryofneurologicalfunction,andprotectsneuralcellsagainstinjury.
