简介:目的比较雌激素对C57BL/6同种系和异种系妊娠小鼠肝脏的影响,探讨妊娠期肝损伤小鼠模型选择C57BL/6异种系妊娠小鼠的可能性。方法选择健康C57BL/6雄鼠、BALB/c雄鼠分别和C57BIM6雌鼠交配,得到C57BL/6同/异种系妊娠小鼠,并分为对照组、溶剂组和不同剂量给药组。给药组从怀孕10d起连续4d分别每天皮下注射不同剂量17-α-乙炔雌二醇。结果给药的异种系孕鼠血清中除碱性磷酸酶(ALP)指标外丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、总胆红素(TBm)和甘胆酸(CG)升高水平与同种系孕鼠比较差异有统计学意义(P〈0.05),其肝脏组织病理学结果也不同,且剂量为1.0μg/g体重的异种系孕鼠肝脏NK细胞出现集聚和活化现象。结论雌激素对C57BIM6同种系和异种系妊娠小鼠肝脏作用不同,选择雌激素诱导C57BL/6异种系妊娠小鼠建立妊娠期肝损伤模型能够反映该病的肝脏免疫学变化。
简介:Theworkistoexploretherelationshipbetweenthelevelsofcytokines(IL-1βandIL-6)inC57BL/6JmicetreatedwithMPTPandbrainlateralization.Byusingpawpreferencetest,right-pawed,left-pawedmicemodelswereestablished.Followingsingleinjectionof1-methyl-4-pheny1-2,3,6-tetrahydropyrid(MPTP)(40mg/kg)toimpairdopaminergicneuron,enzymelinkedimmunosorbentassay(ELISA)kitswereusedfordetectionofplasmalevelsofcytokines.TheresultsshowedthatinsalinetreatedC57BL/6Jmice(control),therewasnoobviousdifferenceobservedbetweenleft-pawedandright-pawedmiceinplasmalevelsofIL-1βandIL-6.InMPTPtreatedmice,therewasnodifferencebetweenlevelofIL-1βinleft-pawedmiceandthatinright-pawedonesinstatistics,thatis,theywereincreasedonday1andday3,butdecreasedonday6.TheplasmalevelofIL-6waslowerinleft-pawedthanthatinright-pawedmice(p<0.005)afterMPTPtreatment.Onday1andday3,thelevelofIL-6wasalmostthesameascontrol;onday6,itwassignificantlyincreased,higherthanthatofcontrol(p<0.001)inleft-pawedmice.Whileinright-pawedmice,onday1andday3,itwasnodifferentfromcontrol,too.Andonday6,itsignificantlyincreasedincomparedwithcontrol(p<0.005).Inconclusion,thelevelofplasmaIL-6ofC57BL/6JmicetreatedwithMPTPincreased.ThevariationofIL-6wascorrelatedtobrainlateralization.
简介:目的探索BALB/c和C57BL/6两个品系在有关实验中的不同作用。方法选取了结合随机测序与生物信息学分析设计合成的神经系统表达的一些基因的反义核酸(antisense)中的2个,用Hamilton微量注射器将其分别定量注射到BALB/c和C57BL/6小鼠的侧脑室,并分别设注射生理盐水和随机序列核酸(Scramble)的对照组。每一反义核酸实验组和对照组各注射10只小鼠,之后观察实验组与对照组在不同行为学实验中的差异。小鼠的行为学检测模型为:考察日常代谢能力的摄食量,考察Locomotionactivity(移动)的旷场行为,考察疼痛阈值的甩尾试验和考察记忆能力的步下法实验。结果注射No.1基因的反义核酸后,两品系的实验组均在测试记忆力的步下法(Step-downTest)试验中表现出记忆力减弱,且与对照组差异明显,说明No.1基因的功能确与记忆相关。注射No.2基因的反义核酸后,在测试移动能力的旷场行为(OpenFieldBehavior)试验中,BALB/c实验组跨格、直立行为均比对照组明显减少,说明受此反义核酸影响显著,而C57BL/6实验组则与对照组无大的差异。此外,在生理盐水对照组和随机序列核酸对照组的实验中以及其他行为学模型的实验中,两品系也存在着一定的差异。结论用遗传背景不同的多品系进行相关实验,可进一步建立新基因功能初筛中有显著结果的基因的复筛平台;同时,实验结果对于进一步研究什么样的品系适用于什么样的实验将具有较大的意义。

简介:Objective:Toexploretheprobabilityofvascularendothelialgrowthfactor(VEGF)antisenseoligodeoxynucleotidesasadevelopingnewtherapeuticstrategyforglioma.Methods:VEGFproteinexpressionwasdetectedbyS-Pimmunohistochemicaltechnique.TumorcellapoptosiswasobservedbyTUNELmethod.Results:Comparedwithcontrol,VEGFproteinexpressionwasinhibitedbyantisenseoligodeoxynucleotidesinvitro.Andtheinhibitoryeffectsincreasedwiththeincreasingconcentration.VEGFpositiveratewas82.10%incontrolgroup,whilein2.5,5,10(mol/LAODNgroups,theywere70.00%,57.85%,53.20%respectively.Noinhibitioneffectwasfoundinthecelllinestreatedwithmissenseandsenseoligodeoxynucleotides.Invivo,antisenseoligodeoxy-nucleotidestherapyalsoinhibitedVEGFproteinexpressionandinducedtheincreaseofapoptotictumorcells.However,ithasnoeffectontumorcellproliferation.Conclusion:ItishopefulthatVEGFantisenseoligodeoxynucleotidesmaybeanewgenetherapymethodtogliomathroughitsantiangiogenesiseffectbyinhibitionofVEGF.
简介:目的:研究外源性C2-神经酰胺对大鼠脑胶质瘤细胞C6活力的抑制作用以及早期凋亡诱导作用.方法:采用MTT法测定C2-神经酰胺对C6细胞活力的影响;倒置荧光显微系统观察细胞形态变化;AnnexinⅤ/PI双染色流式细胞术对细胞早期凋亡进行定量分析.结果:C2-神经酰胺可显著抑制C6细胞的活力,IC50为2.2×10-5mol·L-1;荧光染色可见核浓染等细胞凋亡特征形态改变;流式细胞术分析表明C2-神经酰胺可诱导C6细胞发生早期凋亡作用,且凋亡百分率呈时间及浓度依赖性,C2-神经酰胺2×10-5mol·L-1处理24h后平均早期凋亡率高达49.3%.结论:C2-神经酰胺主要通过诱导早期细胞凋亡对大鼠脑胶质瘤C6细胞发挥细胞毒作用,提高神经酰胺水平有望成为肿瘤化疗的新途径.
