简介:摘要目的探讨微小RNA-642a-5p(miR-642a-5p)对结肠癌细胞株SW620细胞迁移和侵袭的影响及分子机制。方法将人结肠癌细胞SW620细胞分成4个组:对照组(OE-NC+mimic-NC组:共转染miR-642a-5p mimics阴性对照与空载体);过表达miR-642a-5p组(OE-NC+mimic-miR-642a-5p组:共转染miR-642a-5p mimics与空载体);过表达miR-642a-5p与Ⅰ型胶原α1(COL1A1)组(OE-COL1A1+mimic-miR-642a-5p组:共转染miR-642a-5p mimics与COL1A1过表达载体);过表达COL1A1组(OE-COL1A1+mimic-NC组:共转染miR-642a-5p mimics阴性对照与COL1A1过表达载体)。反转录-聚合酶链反应(RT-PCR)检测各组细胞内miR-642a-5p及COL1A1的表达;划痕实验、Transwell小室实验检测细胞迁移和侵袭能力;蛋白质印迹法(Western blot)实验检测细胞内COL1A1蛋白表达。两组间比较采用独立样本t检验。结果RT-PCR结果显示应用miR-642a-5p mimics转染细胞后,miR-642a-5p表达水平显著增加;双荧光素酶报告基因实验结果miR-642a-5p直接靶向作用于COL1A1。应用(COL1A1 overexpression,OE-COL1A1)转染后,细胞COL1A1表达水平明显增加。而OE-NC + mimic-miR-642a-5p组和OE-COL1A1 + mimic-miR-642a-5p组的RNA相对表达量(18.62±2.31),(2.52±0.44)明显高于OE-NC+mimic-NC组(0.52±0.11),差异有统计学意义(t=36.120、6.125,P<0.05),且OE-COL1A1 + mimic-miR-642a-5p组的COL1A1相对表达量(5.88±1.06)明显高于OE-NC+mimic-miR-642a-5p组(0.22±0.04),差异有统计学意义(t=10.201,P<0.05)。与OE-NC + mimic-NC组比较,miR-642a-5p的过表达显著抑制了细胞的迁移和侵袭能力,而过表达COL1A1显著促进SW620细胞的迁移和侵袭能力,并部分逆转miR-642a-5p对细胞迁移和侵袭的抑制作用。结论miR-642a-5p可通过调节COL1A1的表达水平来抑制结肠癌细胞的迁移与侵袭。
简介:摘要目的探讨不同克隆号p53抗体在胃腺癌中的表达及意义,与TP53基因突变情况进行一致性分析,筛选最优克隆号及其阳性阈值。方法收集河南省人民医院病理科2015年6月至2016年12月间42例胃腺癌组织,采用免疫组织化学EnVision二步法分别检测4种不同克隆号(DO-7、BP-53-12、MX008、SP5)p53蛋白水平表达情况,并采用二代测序技术检测TP53基因突变情况。统计分析不同梯度蛋白表达情况,比较各克隆号p53抗体的灵敏度、特异度和准确性。结果4种克隆号(DO-7、BP-53-12、MX008和SP5)p53免疫组织化学染色阳性腺癌细胞百分比各不相同,阳性例数亦不相同,分别为39/42(92.9%)、40/42(95.2%)、36/42(85.7%)和37/42(88.1%)。二代测序TP53基因突变33例,无突变9例。克隆号DO-7在阳性阈值为50%时准确率最高,BP-53-12、MX008和SP5准确率最高的阳性阈值分别为70%、60%和20%。相对应的4种克隆号灵敏度为:DO-7 100.0%、BP-53-12 95.7%、MX008 95.7%及SP5 100.0%;特异度为:DO-7 77.8%、BP-53-12 88.9%、MX008 100.0%及SP5 100.0%。结论不同克隆号p53在胃腺癌蛋白水平表达情况不同,最佳阳性阈值差异较大,评估其免疫组织化学染色结果必须明确所用抗体的克隆号。本研究结果表明,胃腺癌中p53克隆号SP5蛋白水平免疫标记最佳阈值为20%,与TP53基因水平预测的准确率、灵敏度、特异度均达到100.0%,因此,胃腺癌中p53采用克隆号SP5免疫组织化学探讨TP53基因突变背后的机制及创新治疗方式更具有代表性意义和价值。

简介:AbstractForecasting the COVID-19 confirmed cases, deaths, and recoveries demands time to know the severity of the novel coronavirus. This research aims to predict all types of COVID-19 cases (verified people, deaths, and recoveries) from the deadliest 3rd wave data of the COVID-19 pandemic in Bangladesh. We used the official website of the Directorate General of Health Services as our data source. To identify and predict the upcoming trends of the COVID-19 situation of Bangladesh, we fit the Auto-Regressive Integrated Moving Average (ARIMA) model on the data from Mar. 01, 2021 to Jul. 31, 2021. The finding of the ARIMA model (forecast model) reveals that infected, deaths, and recoveries number will have experienced exponential growth in Bangladesh to October 2021. Our model reports that confirmed cases and deaths will escalate by four times, and the recoveries will improve by five times at a later point in October 2021 if the trend of the three scenarios of COVID-19 from March to July lasts. The prediction of the COVID-19 scenario for the next three months is very frightening in Bangladesh, so the strategic planner and field-level personnel need to search for suitable policies and strategies and adopt these for controlling the mass transmission of the virus.
