Phagocytosis of IgA Immune Complexes by Human U937 Cells

摘要 InordertostudyFcαRⅠmediatedphagocytosisotlgAimmunecomplexesbyU937cells,antigen8.9NIP/BSAwaslabeledwithFITCandreactedwithanti-NIPIgAoranti-NIPIgGantibodytoformimmunecomplexes(ICs).Theywerethenincubatedwithphorbol12-myristateB-acetate(PMA)stimulatedU937cells.ThephagocytosedICswerequantifiedbyflowcytometry.TheresultswasthattheexpressionofFcαRⅠonU937cellswashigherthanthatofFcγRⅠ,FcyRⅡandFcγRⅢ.AfterstimulationbyPMA,expressionofFcαRⅠonU937cellswasmarkedlyupregulatedandthephagocytosisofIgAICswasenhanced.FcαRⅠmediatedspecificIgAphagocytosiswasstrongerthanFcγRⅠandFcγRⅡmediatedIgGphagocytosis.Complementreceptors,CR1andCR3,enhancedU937cellphagocytosisofIgAICs.ItconcludesthatFcγRⅠmediatedstrongphagocytosisofIgAICs.
