摘要
ObjectiveTheaimofthisstudyistoinvestigatetheeffectoftransfectedhTERTgeneoncellapoptosisofnewbornratcochlearbasilarmembranecells(CBMCs).MethodsCBMCsisolatedfromnewbornratcochlearweretransfectedusingaplasmidcontaininghumantelomerasereverasetranscriptasegene(pCI-neo-hTERT),andwerescreenedusingG418toobtainstabletransfectedcelllines.Cellapoptosisratewasanalyzedbyflowcytometry.hTERTandapoptosisrelatedgenesexpressionweredetectedbyreversetranscriptasepolymerasechainreaction(RT-PCR).ResultshTERTgeneexpressionwasdetected72hoursaftergenetransfectionintransfectedcells.TheapoptoticrateoftransfectedCBMCssignificantlyreduced.Expressionofapoptosisrelatedgenescorrespondinglychanged.ConclusionTransfectionofhTERTgeneleadstoreducedapoptosisrateinnewbornratCBMCs.andlowerexpressionofapaf1,Caspase3andBCL2intransfectedcellsascomparedtothatofnormalCBMCs.
