摘要
Quartzcrystalmicrobalance(QCM)andcyclicvoltammetry(CV)wereusedtocharacterizethemonolayerofcytochromec(Cytc),whichwasadsorbedongoldfilmmodifiedwithalkanethiolmixedmonolayer.AdirectcomparisonofproteinsurfacecoveragescalculatedfromQCMandcyclicvoltammetricmeasurementsillustratesthattheratiooftheelectroactiveCytctothetotalsurface-confinedCytcis34%,whichsuggeststhattheorientationisamainfactoraffectingtheelectroactivityofCytc.Moreover,surfaceplasmonresonance(SPR)measurementcombinedwithCV'insitu'wasusedtoinvestigatetheconformationalchangeofCytcintheredoxprocess.Besides,Aunanoparticles(AuNPs)wereadsorbedonthesurfaceofCytc.TheresultindicatesthatAuNPspromoteelectrontransferbetweenCytcandthegoldelectrode,andSPRresultsuggestsAuNPsenhanceSPRsignal.
